SIRT6 is a deacetylase of histone H3 and inhibitors of SIRT6 have been thought as potential agents for treatment of diabetes. Herein we report the discovery of a series of new SIRT6 inhibitors containing the skeleton 1-phenylpiperazine. Among them, compound 5-(4-methylpiperazin-1-yl)-2-nitroaniline (6d) is the most potent one, which showed an IC50 value of 4.93 mu M against SIRT6 in the Fluor de Lys (FDL) assay. It displayed K-D values of 9.76 mu M and 10 mu M in surface plasmon resonance (SPR) and isothermal titration calorimetry (ITC) assays, respectively. In selectivity assay, 6d showed no activity against other members of the HDAC family (SIRT1-3 and HDAC1-11) at concentrations up to 200 mu M. In a mouse model of type 2 diabetes, 6d could significantly increase the level of glucose transporter GLUT-1, thereby reducing blood glucose. Overall, this study provides a promising lead compound for subsequent drug discovery targeting SIRT6.

We report the synthesis of alpha-aminonitriles by one-pot coupling reaction of aldehyde, amines and trimethylsilyl cyanide (TMSCN) using beta-ketoiminato zinc complexes as a pre-catalyst in very good yield under mild reaction condition. Homoleptic zinc complex [{kappa(2)-(2,4,6-Me3C6H2NC(Me)=CHC(Me)=O)}(2)Zn] (1 a) was synthesized by the treatment of protic ligand [(2,4,6-Me3C6H2NHC(Me)=CHC(Me)=O)] ((LH)-H-1) with potassium hydride and anhydrous zinc diiodide in 2 : 2 : 1 molar ratio in THF. Analogous reactions using [(2,6-(Pr2C6H3NHC)-Pr-i(Me)=CHC(Me)=O)] ((LH)-H-2) and [(Ph2CHNHC(Me)=CHC(Me)=O)] ((LH)-H-3) as protic ligands, dinuclear zinc complexes [{kappa(3)-(2,6-(Pr2C6H3NC)-Pr-i(Me)=CHC(Me)=O)}ZnI](2)(1 b) and [Zn(Ph2CHNHC-(Me)=CHC(Me)=O)ZnI2] (1 c) were obtained in good yield. Molecular structures of ligandsL(1)H,(LH)-H-3, and zinc complexes1 a,1 b, and1 cwere established by single-crystal x-ray diffraction analysis. Dinuclear zinc complexes1 b, and1 cexhibited high transformation, greater selectivity and broad substrate scope for the synthesis of alpha-aminonitrile under mild condition. A most plausible mechanism for synthesis alpha-aminonitrile is proposed based on several controlled reactions.

Click chemistry is a concept in which modular synthesis is used to rapidly find new molecules with desirable properties(1). Copper(I)-catalysed azide-alkyne cycloaddition (CuAAC) triazole annulation and sulfur(VI) fluoride exchange (SuFEx) catalysis are widely regarded as click reactions(2-4), providing rapid access to their products in yields approaching 100% while being largely orthogonal to other reactions. However, in the case of CuAAC reactions, the availability of azide reagents is limited owing to their potential toxicity and the risk of explosion involved in their preparation. Here we report another reaction to add to the click reaction family: the formation of azides from primary amines, one of the most abundant functional groups(5). The reaction uses just one equivalent of a simple diazotizing species, fluorosulfuryl azide(6-11) (FSO2N3), and enables the preparation of over 1,200 azides on 96-well plates in a safe and practical manner. This reliable transformation is a powerful tool for the CuAAC triazole annulation, the most widely used click reaction at present. This method greatly expands the number of accessible azides and 1,2,3-triazoles and, given the ubiquity of the CuAAC reaction, it should find application in organic synthesis, medicinal chemistry, chemical biology and materials science.

An efficient Ru-catalyzed asymmetric hydrogenation of dibenzo[c,e]azepines is reported. A series of seven-membered cyclic amines were obtained with moderate to excellent enantioselectivity. The catalyst counteranion played an important role in achieving high-level chiral induction. Moreover, a one-pot synthesis of chiral 6,7-dihydro-5H-dibenz[c,e]azepines via two-step reductive amination was also developed.

Construction of N-substituted pyrrolidones from biomass-derived levulinic acid (LA) via reductive amination is a highly attractive route for biomass valorization. However, realizing this transformation using H-2 as the hydrogen source under mild conditions is still very challenging. Herein, we designed porous TiO2 nanosheets-supported Pt nanoparticles (Pt/P-TiO2) as the heterogeneous catalyst. The prepared Pt/P-TiO2 was highly efficient for reductive amination of LA to produce various N-substituted pyrrolidones (34 examples) at ambient temperature and H-2 pressure. Meanwhile, Pt/P-TiO2 showed good applicability for reductive amination of levulinic esters, 4-acetylbutyric acid, 2-acetylbenzoic acid, and 2-carboxybenzaldehyde. Systematic studies indicated that the strong acidity of P-TiO2 and the lower electron density of the Pt sites as well as the porous structure resulted in the excellent activity of Pt/P-TiO2.

Three novel hexacoordinated nickel catalysts containing salicylaldbenzhydrylimine ligand and tetrahydrofuran heterocycle, Ni{(THF) (3-R-1) (5-R-2)C6H2(O)CHNCH(C6H5)(2)}(2), (R-1 = Br, R-2 = H, Ni1; R-1 = Br, R-2 = Br, Ni2; R-1 = Cl, R-2 = Cl, Ni3.), were synthesized and their molecular structures were confirmed by X-ray crystallography. Ni1-Ni3 catalysts exhibited high activities (0.53-2.11 x 10(6) g(polymer)/mol(Ni).h) toward norbornene homopolymerization and high activity (0.29-2.62 x 10(5) g(polymer)/mol(Ni).h) to copolymerization of norbornene and 1-hexene. When the 1-hexene feed ratio was changed from 10 to 70%, the 1-hexene content in the obtained copolymers could be controlled to 8.23-26.58%. The Ni3 with R-1 and R-2 were both electron withdrawing groups (-Cl) on the aromatic ring of salicylaldehyde, showed the highest activity as well as the highest 1-hexene insert ratios. At the same time, only a small amount of B(C6F5)(3) cocatalyst (5 x 10(-5) mol) need to be consumed in the copolymerization process. In addition, the obtained copolymers had a high thermal decomposition temperature (T-d > 386 degrees C) and a low glass transition temperature (T-g = 208.67-237.42 degrees C), which showed that the processability of these polymers was improved while the high thermal decomposition temperature was kept.
