The authors investigated scavenging of superoxide radicals by selenoureas and thioureas using a highly sensitive and quant. chemiluminescence method. At 330 nM, five selenoureas and five thioureas scavenged fractions of superoxide radicals (O2) ranging from 8.4% to 87.6%. Among five N,N-unsubstituted selenoureas and N,N-unsubstituted thioureas, 1-selenocarbamoylpiperidine and 1-thiocarbamoylpyrrolidine were the most effective scavengers. The study showed that selenoureas could be used as new superoxide anion-scavenging substances.

A wide variety of N-substituted 7-amino coumarin analogs was prepared in good to excellent yields. The photophys. properties of aqueous-soluble derivatives were determined, and they displayed auxochrome-based variations. Gram-scale synthesis provided an acrylamide analog, which was used to fabricate a fluorescent poly(2-hydroxylethyl methacrylate) (pHEMA) hydrogel that was resistant to leaching in ultrapure H2O. We envisage that our reported protocol to access 7-amino-4-methylcoumarin derivatives will find use toward the development of new fluorescent coumarin-based probes by researchers in the field.

Starting from thiourea derivatives, a new synthetic pathway to BTZs was established. It allows the formation of the thiazinone ring system in one synthetic step and was applicable for preparation of a wide variety of BTZ analogs. The synthetic procedure furthermore facilitates the replacement of the sulfur atom in the thiazinone ring system by oxygen or nitrogen to afford the analogous benzoxazinone and quinazolinone systems. 36 BTZ analogs were prepared and tested in luminescence-based assays for in vitro activity against Mycobacterium tuberculosis (Mtb) using the microdilution broth method and a high-throughput macrophage infection assay.

Linear free energy relations (LFER) were applied to the IR, 1H- and 13C-NMR spectral data in N-alkyl and N-cycloalkyl cyanoacetamides. N-alkyl and N-cycloalkyl cyanoacetamides were synthesized from corresponding amine and Et cyanoacetate. A number of substituents were employed for alkyl substitution, and fairly good correlations were obtained, using simple Hammett equation. In N-alkyl and N-cycloalkyl cyanoacetamides substituent cause SCS of N-H hydrogen primarily by steric interaction, polar substituent effect influences SCS shift of C=O carbon, while steric effect of N-alkyl substituent causes IR stretching frequencies of N-H, C=O and CN group. The conformations of studied compounds were studied using semiempirical PM6 method, and together with LFER anal., give a better insight into the influence of such a structure on the transmission of electronic substituent effects. Neg. ρ values for several correlations (reverse substituent effect) were found.

Based on in silico approach to screen ascorbic acid as a coformer for curcumin, cocrystals were prepared by the solvent evaporation method, and further evaluated for saturation solubility, cocrystal propensity, physicochem. interactions (FTIR and DSC), XRD, drug dissolution, etc. In silico findings confirmed the suitability (H_ex, G_mix) of ascorbic acid for the cocrystn. of curcumin. The DSC and XRD data of the solvent evaporated curcumin-ascorbic acid mixture confirmed the formation of cocrystal, eutectic, and binary mixture with an excess of coformer. The binary phase diagram implied 0.5 to the 0.65-mol fraction of curcumin, essential for cocrystn. with ascorbic acid. The novel curcumin ascorbic acid cocrystals revealed extraordinary improvement in aqueous solubility of curcumin, especially, 576 fold in distilled water, 10 fold in the buffer pH 1.2, and 9 fold in the buffer pH 6.8. The curcumin-ascorbic acid cocrystal system exhibited a superior dissolution profile compared to neat curcumin. Thus, ascorbic acid has enunciated its role as a coformer for curcumin in cocrystal formation, which has been complemented by predicted complementary biol. activities, and stability (acidic milieu).
