Two kinds of thermo- and photo-dual-responsive polysiloxanes (DRPSs) with functional pendent groups, N-isopropyl amides and azobenzene (Azo) or salicylideneaniline (SA), were synthesized through a facile, effective, and catalyst-free aza-Michael addition of poly(aminopropylmethyl-siloxane) with N-isopropyl acrylamide and N-azobenzene acrylamide or N-salicylaldehyde acrylamide. The chemical structrures of DRPSs were systematically characterized using FT-IR, H NMR and UV-Vis spectroscopy. The as-prepared DRPSs with lower Azo or SA contents exhibited lower critical solution temperature (LCST)-type phase transition in water, which is reversible and can be controlled by temperature and UV light. The effects of Azo and SA contents on the responsive properties of DRPSs are examined in detail. The LCST decreased with the increasing Azo or SA content. Once the content of Azo or SA reached up to 5.7% or 8.2%, respectively, DRPSs could not be dissolved in water even in an ice bath. Higher values of the LCST were measured after irradiation of the polymer solutions due to the higher polarity of cis-Azo and keto-SA conformation, induced by irradiation. The differences in cloud points between the irradiated and the non-irradiated DRPS aqueous solutions increased up to 3.4 degrees C and 9.8 degrees C when combined with 3.8% Azo and 5.8% SA units, respectively.

Post-translational modification of the adeno-associated virus capsids is a poorly understood factor in the development of these viral vectors into pharmaceutical products. Here we report the extensive capsid deamidation of adeno-associated virus serotype 8 and seven other diverse adeno-associated virus serotypes, with supporting evidence from structural, biochemical, and mass spectrometry approaches. The extent of deamidation at each site depended on the vector’s age and multiple primary-sequence and three-dimensional structural factors. However, the extent of deamidation was largely independent of the vector recovery and purification conditions. We demonstrate the potential for deamidation to impact transduction activity and, moreover, correlate an early time point loss in vector activity to rapidly progressing spontaneous deamidation at several adeno-associated virus 8 asparagines. We explore mutational strategies that stabilize side-chain amides, improving vector transduction and reducing the lot-to-lot molecular variability that presents a key concern in biologics manufacturing. This study illuminates a previously unknown aspect of adeno-associated virus capsid heterogeneity and highlights its importance in the development of these vectors for gene therapy.

Owing to its high sensitivity, surface-enhanced Raman spectroscopy (SERS) is widely used in biosensing as an efficient optical transduction technique. In this work, we report the elaboration of a novel SERS-powered aptasensing substrate for the ultra-sensitive and selective detection of prostate specific antigen (PSA). The transducing SERS substrate is formed by vertically aligned silicon nanowires (SiNWs) decorated with silver nanoparticules (AgNPs). The silver nanoparticles have two purposes : a) aptamer immobilization via thiol chemistry and b) Raman signal enhancement. The presence of PSA induces the appearance of amide bond vibration modes only if the aptamer is present on the surface of SiNWs. The platform is highly sensitive, selective and specific to PSA in a wide range of concentrations from 0.1 to 20 mu g.L-1 with a detection limit of 0.1 mu g.L-1, which encompasses the blood serum range of healthy subjects and ill patients. This optical biosensor shows a great stability and it is successfully validated in human blood serum samples showing very good results.

The Ni2+ complexes with hexadentate ligands containing two 6-methylpicolinamide groups linked by ethane-1,2-diamine (dedpam) or cyclohexane-1,2-diamine (chxdedpam) spacers were investigated as potential contrast agents in magnetic resonance imaging (MRI). The properties of the complexes were compared to that of the analogues containing 6-methylpicolinate units (dedpa(2-) and chxdedpa(2-)). The protonation constants of the dedpa(2-) and dedpam ligands and the stability constants of their Ni2+ complexes were determined using pH-potentiometry and spectrophotometric titrations (25 degrees C, 0.15 M NaCl). The [Ni(dedpa)] complex (log K-NiL = 20.88(1)) was found to be considerably more stable than the corresponding amide derivative [Ni(dedpam)](2+) (log K-NiL = 14.29(2)). However, the amide derivative [Ni(chxdedpam)](2+) was found to be considerably more inert with respect to proton-assisted dissociation than the carboxylate derivative [Ni(chxdedpa)]. A detailed H-1 NMR and DFT study was conducted to assign the H-1 NMR spectra of the [Ni(chxdedpa)] and [Ni(chxdedpam)](2+) complexes. The observed H-1 NMR paramagnetic shifts were found to be dominated by the Fermi contact contribution. The amide resonances of [Ni(chxdedpam)](2+) at 91.5 and 22.2 ppm were found to provide a sizeable chemical exchange saturation transfer effect, paving the way for the development of NiCEST agents based on these rigid non-macrocyclic platforms.

The wet solid was taken into another 100 mL round bottom flask and n-butylacetate (50 mL) was added and stirred for 30 minutes at 20C. The precipitation was filtered and the solid was suck dried. The material was dried at 45 under vacuum for 5 hours to yield 14 gm of title compound.

Fractions containing the desired product were combined and dried via centrifugal evaporation. The yield of the product (Example 29) was 15.4 mg (49%). HPLC (Method E) RT = 0.98 min. HPLC (Method G) RT = 0.76 min. The hydrobromide salt is dissolved in DMF (8mL) together with diisoproylethylamine (0.38 mL, 2.13 mmol), i-hydroxybenzotriazole (107 mg, 0.71 mmol) and (5)-(2-methoxycarbonyl-amino)-3-phenylpropionic acid (175 mg, 0.78 mmol). The mixture is stirred at 0 C for 30 minutes then at room temperature overnight. The reaction mixture is diluted with water and extracted with EtOAc. The combined organic phase is washed with 1 N aqueous HC1, 5 % aqueous NaHCO3, water and brine, and dried over Na2SO4. The solvent is removed in vacuo to afford 0.300g (81% yield) of the desired product which is used without further purification. LC/MSESI+MS 483 (M+1).

The reaction mixture was warmed up to room temperature and stirred overnight. The solution was concentrated and the residue was diluted with ethyl acetate, washed consecutively with 1N HCl and water, dried with Na2SO4, filtered and evaporated to afford a colorless oil. The oil was refluxed in hexane for 1 hour then cooled to room temperature and filtered to afford the target compound as a white solid (12.1 g, 42.1% yield). 1H NMR (DMSO-d6, 300 MHz) delta ppm 11.22 (s, 1H), 3.18 (s, 3H), 1.42 (s, 9H).
