Highly real nanostructured clay adsorbents of layered double hydroxide (LDH) intercalated with biosafe and anionic organic diacid was produced at a constant pH with the coprecipitation method. XRD results disclosed that organic anionic segment is horizontally oriented in the interlayer space, which is obvious from developed basal spacing of 0.76 to 1.36 nm. Also, a successfully LDH intercalation with examined biosafe diacid was observed using FT-IR and TGA analysis. Organo-intercalated LDH (I-LDH)/PVC nanocomposites (NCs) with different content of I-LDH loading were organized through ultrasound irradiation as a green method. The morphological behaviors and structured properties of achieved NCs were studied by characteristic analyses such as FT-IR, XRD FE-SEM and TEM. Results of thermal analysis disclosed positive influence of I-LDH on degradation temperature in compared to the neat PVC, and also residue at 800 degrees C enhanced from 23 to 42%. The resulting LDH samples were studied for their performance in removing Cd2+ metal ion from aqueous test solution and the removal efficiency of these materials at proper pH, different adsorbent mass, Cd2+ concentrations and different times were documented by atomic adsorption spectrometry. In all samples, with the developed in the adsorbent mass, the removal efficiency for Cd2+ was improved at the 50 mg/L of metal ion concentration and constant pH of 7.0. The maximum Cd2+ uptake was 86.17% related to the I-LDH/PVC NC 15 wt%, and also the experimental data indicated a good compliance with the Langmuir isotherm and pseudo-second-order kinetic model for different LDH samples.

The reaction has the advantages of mild conditions, less by-products, good yield and simple post-treatment, and is a new method for the efficient preparation of 3-hydroxy-3-heterocycle butylamides.

Improvement of the Antioxidant Activity of Fenugreek Protein Isolates by Lactococcus lactis Fermentation

This study investigated the antioxidant capacity of fenugreek protein isolate and its improvement by Lc. lactis fermentation through bioactive peptides production and the effect of molecular weight variation on fenugreek fractions antioxidant activity. Fenugreek protein isolate showed a significant increase of antioxidant and radical scavenging activity after 24h of fermentation, by about 23.7, 42.9 and 40% for respectively antioxidant activity coefficient AAC, DPPH and ABTS radical scavenging activity. FI fermentation led to a hydrolysis of peptide bands with MW>35kDa and a generation of new bands with a MW<25kDa. A significant reduction in molecular-mass distribution of hydrolysates and a great increase of total free amino acids content, especially an increase on isoleucine, leucine, glutamic acid, serine, histidine, glutamine and lysine was noted. The infrared results showed that different reactions may take place after fermentation, and showed an increase of proteins, amides and aromatic compounds. However, fenugreek fraction (F2) with MW 15-50kDa presented the highest activity instead of fraction (F1) with lower MW. Lc. lactis had the ability to degrade and convert fenugreek proteins into bioactive peptides that contribute positively in the improvement of antioxidant activity of FI and fractions. FI presents a significant antioxidant activity and thus, can be considered as a potential source of high added value natural antioxidants and may be employed as a functional food ingredient with good potential applications in food products. Development and validation of a bioanalytical method of analyzing 3 ‘- and 6 ‘-sialyllactose using liquid chromatography-tandem mass spectrometry in minipig plasma and its application in a pharmacokinetic study

Sialyllactose (SL) is an acidic oligosaccharide, consisting of a combination of sialic acid and lactose. It is found in human milk. It has immune-protective effects against pathogens in newborns and helps with the development of the immune system and intestinal microorganisms. We developed and validated a method by which 3′-SL and 6′-SL levels were simultaneously analyzed via liquid chromatography-tandem mass spectrometry (LC-MS/MS), and evaluated the pharmacokinetics of the materials after systemic delivery to minipigs. To improve chromatographic selectivity, several types of columns (C18, amide, and HILIC phase) were used to separate the peaks of 3′-SL and 6′-SL. Ultimately the HILIC phase column was selected, as it had a good peak shape and quick resolution. The mobile phase comprised ammonium acetate buffer and acetonitrile with gradient elution. MS was performed in the negative ion and multiple reaction monitoring modes. Plasma samples were prepared using the protein precipitation method with methanol. A surrogate matrix was used for quantification because SLs are endogenous plasma compounds. The method developed was validated according to U.S. Food and Drug Administration guidance. A pharmacokinetic study was performed with intravenous administration of 3′-SL and 6′-SL in minipigs (Sus scrofa/Yucatan). The concentrations of 3′-SL and 6’-SL were readily measurable in the plasma samples, which suggests that the method adequately determined systemic exposure in minipigs. (C) 2020 Elsevier B.V. Antibacterial Natural Peptide Fractions from Indian Ganoderma lucidum

Natural peptides are emerging as a leading alternative to conventional drugs and antibiotics, owing to their remarkable potency, better stability and lesstoxicity. Such peptides encompass numerous healing properties such as antimicrobial, anti-inflammatory,immunomodulatory, etc. Though plant- derived peptides have been widely studied for their therapeutic benefits, however, fungal peptides are still lesser explored. Ganoderma lucidum, a highly medicinal oriental mushroom comprises a vast array of phytoconstituents, namely flavonoids, phenolics, terpenoids, polysaccharides, proteins, glycolipids, etc and hence, is being used since several decades in traditional Chinese medicine (TCM) for its various ameliorative effects e.g. anti-inflammatory, antimicrobial, anti-proliferativeandantioxidantproperties. This study presents the isolation and characterization ofantibacterial peptide fractions from fruiting body (GLF) and mycelium (GLM) of Indian G. lucidum. Representative amide bonds were identified in the fractions using established standard techniques. Peptide mass fingerprinting and HPLC confirmed the presence of cationic and hydrophobic amino acids in the peptide fractions which are known to be major structural features of antimicrobial peptides. Secondary structure prediction showed abundance of -helices and random coils in GLF and GLM fractionsrespectively. The fractions exhibited appreciable antioxidant potential. Besides, these also possessed substantial antibacterial activity against Escherichia coli and Salmonella typhi wherein it was observed that generation of reactive oxygen species and induction of intracellular protein leakage within the bacterial cells were the possible mechanisms of inhibitory action.

Lower amygdala fatty acid amide hydrolase in violent offenders with antisocial personality disorder: an [C-11]CURB positron emission tomography study

Antisocial personality disorder (ASPD) imposes a high societal burden given the repetitive reactive aggression that affected individuals perpetrate. Since the brain endocannabinoid system (ECS) has been implicated in ASPD and aggressive behavior, we utilized [C-11]CURB positron emission tomography to investigate fatty acid amide hydrolase (FAAH), an enzyme of the ECS that degrades anandamide, in 16 individuals with ASPD and 16 control participants. We hypothesized that FAAH density would be lower in the amygdala for several reasons. First, decreased FAAH expression is associated with increased cannabinoid receptor 1 stimulation, which may be responsible for amygdala hyper-reactivity in reactive aggression. Second, the amygdala is the seat of the neural circuit mediating reactive aggression. Third, other PET studies of externalizing populations show reduced brain FAAH density. Conversely, we hypothesized that FAAH expression would be greater in the orbitofrontal cortex. Consistent with our hypothesis, we found that amygdala FAAH density was lower in the amygdala of ASPD (p=0.013). Cerebellar and striatal FAAH expression were inversely related with impulsivity (cerebellum: r=-0.60, p=0.017; dorsal caudate: r=-0.58, p=0.023; dorsal putamen: r=-0.55, p=0.034), while cerebellar FAAH density was also negatively associated with assaultive aggression (r=-0.54, p=0.035). ASPD presents high levels of disruptive behavior with few, if any, efficacious treatment options. Novel therapeutics that increase FAAH brain levels in a region-specific manner could hold promise for attenuating certain symptom clusters of ASPD, although our results require replication.

Effect of cannabidiol on endocannabinoid, glutamatergic and GABAergic signalling markers in male offspring of a maternal immune activation (poly I:C) model relevant to schizophrenia

The mainstay treatment for schizophrenia is antipsychotic drugs (APDs), which are mostly effective against the positive symptoms (e.g. hallucinations), but provide minimal benefits for the negative symptoms (e.g. social withdrawal) and cognitive deficits. We have recently shown that treatment with the non-intoxicating phyto-cannabinoid, cannabidiol (CBD), can improve cognition and social interaction deficits in a maternal immune activation (MIA) model relevant to the aetiology of schizophrenia, however, the mechanisms underlying this effect are unknown. An imbalance in the main excitatory (glutamate) and inhibitory (GABA) neurotransmitter systems in the brain plays a role in the pathophysiology of schizophrenia. Therefore, the endocannabinoid system could represent a therapeutic target for schizophrenia as a regulator of glutamate and GABA release via the CB1 receptor (CB1R). This study investigated the effects of chronic CBD treatment on markers of glutamatergic, GABAergic and endocannabinoid signalling in brain regions implicated in social behaviour and cognitive function, including the prefrontal cortex (PFC) and hippocampus (HPC). Time-mated pregnant Sprague-Dawley rats (n = 16) were administered poly I:C (4 mg/kg, i.v.) or saline (control) on gestational day 15. Male offspring were injected with CBD (10 mg/kg, i.p.) or vehicle twice daily from postnatal day 56 for 3 weeks. The prefrontal cortex (PFC) and hippocampus (HPC) were collected for post-mortem receptor binding and Western blot analyses (n = 8 per group). CBD treatment attenuated poly I:C-induced deficits in cannabinoid CB1 receptor binding in the PFC and glutamate decarboxylase 67, the enzyme that converts glutamate to GABA, in the HPC. CBD treatment increased parvalbumin levels in the HPC, regardless of whether offspring were exposed to poly I:C in utero. Conversely, CBD did not affect N-methyl-D-aspartate receptor and gamma-aminobutyric acid (GABA) A receptor binding or protein levels of fatty acid amide hydrolase, the enzyme that degrades the endocannabinoid, anandamide. Overall, these findings show that CBD can restore cannabinoid/GABAergic signalling deficits in regions of the brain implicated in schizophrenia pathophysiology following maternal poly I:C exposure. These findings provide novel evidence for the potential mechanisms underlying the therapeutic effects of CBD treatment in the poly I:C model.

Synthesis of spiropyrrolidine oxindoles via Ag-catalyzed stereo- and regioselective 1,3-dipolar cycloaddition of indole-based azomethine ylides with chalcones

The synthesis of novel spiropyrrolidine oxindole derivatives was reported, using Ag-catalyzed [3+2] cycloaddition of azomethine ylides generated in situ from the condensation of substituted isatins and primary alpha-amino acid esters with chalcones. Products bearing four consecutive stereocenters, including spiroquaternary stereocenters fused in one ring structure, were smoothly acquired in moderate to high yields (50-95%) with good to excellent diastereoselectivities (11 : 1 -> 20 : 1 dr). Furthermore, product 4a underwent reduction, oxidation, hydrolysis and amidization to give the corresponding alcohol, dihydropyrrole, pyrrole, acid and amide, respectively, in good yields. Zirconium-based conversion film formation on zinc, aluminium and magnesium oxides and their interactions with functionalized molecules

Zirconium-based conversion treatment of zinc, aluminium and magnesium oxides have been studied in-situ using ATR-FTIR in a Kretschmann geometry. This set-up was coupled to an electrochemical cell, which allowed to obtain chemical and electrochemical information simultaneously as a function of conversion time. This elucidated the strong relation between physico-chemical surface properties and zirconium-based conversion kinetics. Whereas the surface hydroxyl density of zinc and aluminium increased during conversion, magnesium (hydr) oxide was shown to dissolve in the acid solution. Due to this dissolution, strong surface alkalization can be expected, explaining the rapid conversion kinetics. AES depth profiling was used to determine the final oxide thickness and elemental composition. This confirmed that magnesium is most active and forms a zirconium oxide layer approximately 10 times thicker than zinc. On the other hand, the presence of zirconium oxide on aluminium is very low and can be considered as not fully covering the metal oxide. Additionally, the converted oxide chemistry was related to the bonding mechanisms of amide functionalized molecules using ATR-FTIR and XPS. It was shown that inclusion of zirconium altered the acid-base properties, increasing the substrate proton donating capabilities in case of magnesium oxide and increasing hydrogen bonding and Bronsted interactions due to increased surface hydroxide fractions on zinc and aluminium substrates. Quantitative growth evolution of gold nanoparticles synthesized using aqueous Elaeis guineensis (oil palm) leaves extract

Phytosynthesis of gold nanoparticles (AuNPs) have shown the tremendous interest owing to their ecofriendly, simple, cost effective and renewable features. The growth kinetics behavior of plants mediated AuNPs is relatively scarce despite the significant impacts of their morphological structures on their performance in various fields of sciences. Herein, AuNPs were synthesized employing aqueous Elaeis guineensis leaves extract and characterized using TEM, UV-vis spectroscopy and DLS. Ostwald ripening (OR) and Orientation attachment (OA) models were used to investigate the mechanisms leading towards the growth of AuNPs as function of time. The changes in the positions of UV-vis spectra peaks and increasing number of agglomerated particles as function of time detected the ongoing growth process of AuNPs. The experimental results exhibited that the growth of AuNPs was mainly governed by the OA kinetics in initial phase owing to the stable surface adsorption of phenolic compounds, flavonoids, carboxylic acids and amides, impeding the diffusion-controlled growth. Moreover, TEM analysis also confirmed the domination of OA mechanism in early stage through displaying the formation of AuNPs with quasi spherical and elongated morphology. However, the OA growth kinetics was found to be majorly substituted and followed by the OR mechanism in the later period.
