Synthetic cross-linked collagen-related peptide (CRP-XL) is a glycoprotein VI (GPVI) receptor activator for platelet activation. This triple helical peptide, widely used in platelet function tests, is synthesized and cross-linked through cysteine residues at its N-terminus and C-terminus. Currently, there is only one laboratory, which is capable to produce this valuable peptide for clinical applications. In an attempt to provide a standardized alternative for CRP-XL, we developed a synthetic triple helical collagen peptide (STH-CP) with the same primary sequence as CRP-XL (GPC-(GPO) (10) -GPCG-amide) (3) , which was both on the C-terminus and on the N-terminus fixed on a scaffold with a binding side for each of the three peptides. The performance of STH-CP on platelet function was studied using flow cytometry and compared with CRP-XL. We found that platelet activation pattern in response to STH-CP and CRP-XL is similar, although the STH-CP requires sixfold higher concentrations to activate platelets to the same state. The intra-assay percent coefficient of variation of STH-CP and CRP-XL were both < 5% and the interindividual variation measured in 118 individuals for both peptides was around 23 and 21% for alpha IIb beta 3 activation and P-selectin expression, respectively. The STH-CP in ready-to-use reaction mix has lower variation than CRP-XL over 1-year storage. In reference values and seasonal variation study, the platelet activation response showed a strong correlation between STH-CP and CRP-XL. Our findings show that this new STH-CP is a stable and potent platelet GPVI agonist which can induce the same reproducible platelet activation as CRP-XL and that STH-CP can be considered as a good alternative for CRP-XL. A two-dimensional molybdenum disulfide (MoS2) nanosheet, as a new type of inorganic material with high hydrophobicity and excellent physicochemical stability, holds great application potential in the preparation of a high separation performance organic-inorganic hybrid membrane. In this work, high hydrophobic MoS2 was embedded in hydrophobic polyether copolymer block amide (PEBA) to prepare PEBA/MoS2 organic-inorganic hybrid membranes. The structure, morphology, and hydrophobicity of the hybrid membrane were characterized by scanning electron microscopy, thermogravimetric analysis, contact angle goniometry, X-ray diffraction, infrared spectroscopy analysis, and atomic force microscopy. The effect of embedding of MoS2 on the swelling degree and pervaporation separation performance of the PEBA/MoS2 hybrid membrane was studied with a 1.0 wt % pyridine dilute solution. The results indicated that with increasing the MoS2 content, the separation factor of PEBA/MoS2 increased first and then decreased, while it showed a downward trend in the permeation flux. When the MoS2 content in the PEBA/MoS2 hybrid membrane was 10.0 wt %, the permeation flux was 83.4 g m(-2)h(-1) (decreased by 21.5% compared with the pure PEBA membrane), and the separation factor reached a maximum value of 11.11 (increased by 37.6% compared with the pure PEBA membrane) Meanwhile, the effects of feed temperature on the pervaporation separation performance of PEBA/MoS2 hybrid membranes were also studied. In addition, as the PEBA/MoS2 hybrid membrane has excellent thermal stability, it is expected to be a promising material for recovering pyridine from wastewater.

To explore anticancer and antioxidant agents with improved potency, we synthesized a series of amide linked 1,4-disubstituted 1,2,3-triazoles through click chemistry approach. The antioxidant activity was evaluated by using stable free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay and compound 7d showed promising activity having IC50 value 1.61 mu g/ml. Molecular docking studies of compounds 7e and 7f was carried out in active site of human epidermal growth factor receptor 2 revealed high binding affinities and within toxicity limits. Discovery of a potent p38 alpha/MAPK14 kinase inhibitor: Synthesis, in vitro/in vivo biological evaluation, and docking studies

This article reports the synthesis of new triarylpyrazole derivatives possessing urea or amide linker, and their biological activities at molecular, cellular, and in vivo levels. Compound 2b was the most potent inhibitor of p38 alpha/MAPK14 kinase (IC50 = 22 nM) among this series. Molecular docking studies were conducted to understand the kinase inhibitory variations and the basis of selectivity. Compound 2b was able to inhibit p38 alpha/MAPK14 kinase inside HEK293 cells in nanoBRET cellular kinase assay with EC50 value of 0.55 mu M, comparable to the potency of dasatinib. Compound 2b inhibited TNF-alpha production in lipopolysaccharide-induced THP-1 cells with IC50 value of 58 nM. In addition, compound 2b showed low potency against hERG. It is 62238 times less potent than E-4031 against hERG, so the risk of cardiotoxicity of the compound is very minimal. Compound 2b showed also high plasma stability in vitro in human and rat plasmas.
