Background: A variety of medications are administered to the intra-articular space for the relief of joint pain. While amide-type local anesthetics have been extensively studied, there is minimal information regarding the potential chondrotoxicity of nonsteroidal anti-inflammatory drugs (NSAIDs) and opioid medications. Purpose: To investigate the in vitro chondrotoxicity of single-dose equivalent concentrations of ketorolac, morphine, meperidine, and fentanyl on human chondrocytes. Study Design: Controlled laboratory study. Methods: Human cartilage was arthroscopically harvested from the intercondylar notch and expanded in vitro. Gene expression of cultured chondrocytes before treatment was performed with quantitative polymerase chain reaction for type I collagen, type II collagen, aggrecan, and SOX9. Chondrocytes were then exposed to 0.01%, 0.02%, and 0.04% morphine sulfate; 0.3% and 0.6% ketorolac tromethamine; 0.5%, 1.0%, and 1.5% meperidine hydrochloride; 0.0005% and 0.001% fentanyl citrate; and saline. A custom bioreactor was used to constantly deliver medications, with the dosage of each medication and the duration of exposure based on standard dose equivalents, medication half-lives, and differences in the surface area between the 6-well plates and the native joint surface. After treatment, a live/dead assay was used to assess chondrocyte viability and if minimal cell death was detected. A subset of samples after treatment was maintained to analyze for possible delayed cell death. Results: All tested concentrations of ketorolac and meperidine caused significantly increased cell death versus the saline control, demonstrating a dose-response relationship. The morphine and fentanyl groups did not show increased chondrotoxicity compared with the saline group, even after 2 weeks of additional culture. Conclusion: In vitro exposure of chondrocytes to single-dose equivalent concentrations of either ketorolac or meperidine demonstrated significant chondrotoxicity, while exposure to morphine or fentanyl did not lead to increased cell death.

State-of-the-art perovskite-based solar cells employ expensive, organic hole transporting materials (HTMs) such as Spiro-OMeTAD that, in turn, limits the commercialization of this promising technology. Herein an HTM (EDOT-Amide-TPA) is reported in which a functional amide-based backbone is introduced, which allows this material to be synthesized in a simple condensation reaction with an estimated cost of <$5 g(-1). When employed in perovskite solar cells, EDOT-Amide-TPA demonstrates stabilized power conversion efficiencies up to 20.0% and reproducibly outperforms Spiro-OMeTAD in direct comparisons. Time resolved microwave conductivity measurements indicate that the observed improvement originates from a faster hole injection rate from the perovskite to EDOT-Amide-TPA. Additionally, the devices exhibit an improved lifetime, which is assigned to the coordination of the amide bond to the Li-additive, offering a novel strategy to hamper the migration of additives. Clustering-triggered Emission of Nonaromatic Polymers with Multitype Heteroatoms and Effective Hydrogen Bonding

Nonconventional luminophores without large conjugated structures are attracting increasing attention for their unique aggregation-induced emission(AIE) properties and promising applications in optoelectronic and biomedical areas. The emission mechanism, however, remains elusive, which makes rational molecular design difficult. Recently, we proposed the clustering-triggered emission(CTE) mechanism to illustrate the emission. The clustering of electron-rich nonconventional chromophores with pi and/or n electrons and consequent electron cloud overlap is crucial to the luminescence. Herein, based on the CTE mechanism, nonaromatic polymers containing multitype heteroatoms(i.e., O, N, and S) and involving amide(CONH) and sulfide(-S-) groups were designed and synthesized through facile thiol-ene click chemistry. The resulting polymers demonstrated typical concentration-enhanced emission, AIE phenomenon, and excitation-dependent emission. Notably, compared with polysulfides, these polymers exhibited much higher solid-state emission efficiencies, because of the incorporation of amide units, which contributed to the formation of emissive clusters with highly rigidified conformations through effective hydrogen bonding. Furthermore, distinct persistent cryogenic phosphorescence or even room temperature phosphorescence(RTP) was noticed. These photophysical behaviors can well be rationalized in terms of the CTE mechanism, indicating the feasibility of rational molecular design and luminescence regulation.

Antioxidant functionalization of biomaterials via reactive extrusion

Concern over environmental contamination and the need for industrially produced functional biopolymers motivates the utilization of reactive extrusion for functional grafting. Antioxidant nonmigratory active materials were synthesized through 1,3-bis(4,5-dihydro-2-oxazolyl)benzene ring opening polymerization with bio-based poly(lactic acid) (PLA) and antioxidant nitrilotriacetic acid (NTA). Grafting was confirmed through the introduction of new alkyl stretching, disappearance of the characteristic 1,3-bis(4,5-dihydro-2-oxazolyl)benzene band, and emergence of new amide band on attenuated total reflectance Fourier transform infrared spectroscopy. X-ray photoelectron spectroscopy (XPS) showed stepwise increases in atomic nitrogen percentage and shifts in proportion of CC:CO bonding, confirming grafting and surface orientation. Antioxidant samples exhibited significant increases in carboxylate density, ranging from 0.75 +/- 0.04 to 3.11 +/- 0.04 nmol/cm(2). Functionalized films demonstrated significant antioxidant properties with Trolox((eq)) ranging from 0.36 +/- 0.02 to 0.89 +/- 0.07 nmol/cm(2) according to radical scavenging studies and delayed greater than 52% of ascorbic acid degradation. This work develops a rapid functionalization method for biopolymers and displays efficacy in producing sustainable nonmigratory active materials.

Identification and photostability of N-alkylamides from Acmella oleracea extract

The identification of N-alkylamides from commercial Acmella oleracea extract, their UV-B photostability in different solvents, and identification of degradation products were the main goals of this study. By UHPLC-DAD-ESI-MS/MS method the presence of nine N-alkylamides was identified. Investigation of UV-B irradiation effect on identified N-alkylamides from Acmella oleracea extract was monitored in various the most commonly used solvents (methanol, ethanol, saline solution, and water) during 120 min. The results obtained indicated that spilanthol and homospilanthol were the most stable N-alkylamides presented in Acmella oleracea extract, while the photostability of identified N-alkylamides in whole in tested extract solutions decreased as follows: methanol>ethanol>saline solution>water. As the main degradation products in all investigated solutions 6,9-dihydroxy-deca-2,7-dienoic acid isobutyl-amide and 8,9-dihydroxy-deca-2,6-dienoic acid isobutyl-amide were identified. Deamidation of Amino Acids on the Surface of Adeno-Associated Virus Capsids Leads to Charge Heterogeneity and Altered Vector Function

Post-translational modification of the adeno-associated virus capsids is a poorly understood factor in the development of these viral vectors into pharmaceutical products. Here we report the extensive capsid deamidation of adeno-associated virus serotype 8 and seven other diverse adeno-associated virus serotypes, with supporting evidence from structural, biochemical, and mass spectrometry approaches. The extent of deamidation at each site depended on the vector’s age and multiple primary-sequence and three-dimensional structural factors. However, the extent of deamidation was largely independent of the vector recovery and purification conditions. We demonstrate the potential for deamidation to impact transduction activity and, moreover, correlate an early time point loss in vector activity to rapidly progressing spontaneous deamidation at several adeno-associated virus 8 asparagines. We explore mutational strategies that stabilize side-chain amides, improving vector transduction and reducing the lot-to-lot molecular variability that presents a key concern in biologics manufacturing. This study illuminates a previously unknown aspect of adeno-associated virus capsid heterogeneity and highlights its importance in the development of these vectors for gene therapy.
