Considering the importance of polylactic acid (PLA) biopolymer in industry, in this research we represent a protocol that benefits from the presence of PA6 phase (30 wt%), using an ester-amide exchange catalyst and GN as nanofiller to enhance PLA mechanical properties. To this purpose, it was decided to fabricate blends and nanocomposites in the presence of cobalt(II) acetylacetonate (Cat) as the catalyst. XRD patterns revealed good compatibility of the polymer moieties by broadening of the PLA characteristic peaks and also good dispersion of the GN nanoparticles within the matrix. In the TEM pictures, the GN nanoparticles size ranged from 20 to 130 nm which confirms its good intercalation and exfoliation. T-0 and T-max, considered as thermal stability, of the samples decreased slightly by reactive blending, however, the values were still comparable with them in pure PLA. DSC thermograms and the curves from DMTA exhibited the lower T-g, T-m and crystallinity values of PLA matrix in the alloys and nanocomposites especially in the presence of Cat. Investigation of tensile properties revealed significantly enhanced elongation at break, from 7 in origin PLA to the maximum of 29% in PLA/PA/Cat, with negligible deficiency in modulus, from 2.85 to a minimum of 1.95 GPa, and tensile strength, from 65 to a minimum of 46 MPa, respectively. MFI test results disclosed higher flow-ability of the alloys and composites in compare with neat PLA and also melt flow ratio in two different testing weighs, shows broad molecular weight distribution of the fabricates due to the occurrence of exchange reactions in the presence of Cat.

The placebo effect is the clinical benefit caused by interaction with a caregiver and health care system in the absence of a biologically active intervention and has been used successfully for millennia. The placebo response results from the interaction of psychosocial mechanisms, human relationships, and preconceptions functioning in specific neuroan-atomic locations with known genes and neurotransmitters. It occurs with or without the administration of an inactive substance to deliberately deceive patients. Our purpose is to review the history, benefits, and mechanisms of the placebo effect. The placebo response results from classic conditioning and positive expectations about outcome expressed by the caregiver. The outcomes are usually symptoms such as pain rather than biological outcomes such as death, and the powerful placebo may account for more than half the effect of treatment in many situations. The placebo effect results from activation of opioid, cannabinoid, and dopaminergic pathways involved in reward, expectancy, conditioning, and pain modulation. Eleven specific anatomic features in the brain identified by positron emission tomography and magnetic resonance imaging are involved. Polymorphisms in the structural genes for catecholamine O-methyltransferase and fatty acid amide oxidase significantly influence the placebo response. The placebo effect may be important in symptom suppression in angina, paroxysmal atrial fibrillation, and congestive heart failure. In the absence of deliberate deception, there are no ethical issues and given its potency, the time has come to consider how best to use the placebo in clinical practice.

The bio-crude oil produced from hydrothermal liquefaction (HTL) of a high-protein microalgae useful for wastewater treatment, Galdieria sulphuraria, was comprehensively characterized, and compared to that of a high lipid microalgae useful for biofuel production, Nannochloropsis sauna. HTL was conducted in a batch reactor at temperatures of 310-350 degrees C and reaction times of 5-60 min. Characterization methods included high-resolution Fourier transform ion cyclotron resonance mass spectroscopy (FT-ICR MS), fatty add methyl ester (FAME) analysis by gas chromatography mass spectroscopy (GC/MS), proton nuclear magnetic resonance spectroscopy (H-1 NMR), and Fourier transform infrared spectroscopy (FT-IR). Milder reaction conditions favored bio-crude oil yield and quality for N. sauna, while more severe conditions (350 degrees C) were needed for G. sulphuraria. N. salinaderived bio-crude oil contained mainly C-14-C-18 fatty acid amides, while G. sulphuraria-derived bio-crude-oil had many N1-3O0-2 hetero-atom compounds. FT-ICR MS showed that the aromaticity of hetero-compounds in N. sauna bio-crude oil was higher due to N. sauna’s higher carbohydrate content and the tendency of carbohydrate derived molecules to condense at HTL conditions. FAME-GC/MS and H-1-NMR results showed that stable fatty acid amides increased in G. suiphuraria bio-crude oil at higher temperatures as more protein-derived compounds combined with lipid-derived compounds. While N-containing and high molecular weight compounds are a concern for the upgrading of bio-crude oils obtained from high-protein algal biomass, removal of carbohydrates rather than removal of proteins as a pretreatment to HTL is recommended since carbohydrate-derived compounds are more likely to create highly aromatic hetero-compounds that are much more difficult to upgrade.

The specific interactions between polyacrylamide brushes composed of polyacrylamide chains tethered to montmorillonite particles (Mt-g-PAm) and the individual components of immiscible ethylene-propylene-diene modified with sorbic acid (EPDM) and methyl vinyl silicone (MVQ) blends were studied. The effect of various lengths of PAm chains and their grafting density on montmorillonite (Mt) surface was investigated. It was determined that, due to interactions generated within the system through carboxyl and amide groups the brush particles were selectively located at the EPDM/MVQ interphase and in the EPDM phase. Furthermore, the longer PAm chains resulted in an increase of the glass transition of EPDM, unification of blend morphology, and the reduction of crystallinity of the MVQ phase. Therefore, precisely designed polymer brushes could be considered as novel functional compatibilizers, that provide a new tool for the future material designing.

Hydrozirconation of various C=X functionalities with Schwartz’s reagent is a powerful method for preparing functionalized organic molecules, for instance aldehydes, amines, and amides. The reactions of [Cp2Zr(H)Cl] with amides, lactams, nitriles or isocyanates proceed under mild conditions. These transformations are remarkably chemoselective as Schwartz’s reagent is compatible with various functional groups, including electrophilic functionalities such as ester, nitrile, nitro, and halogens. This compatibility enables many hydrozirconation reactions to be combined with other synthetic methods, and enables performing the reactions in a one-pot fashion as multicomponent processes.

Background: Carboxylesterases (CEs) belong to the serine hydrolase family, and are in charge of hydrolyzing chemicals with carboxylic acid ester and amide functional groups via Ser-His-Glu. Uncaria rhynchophylla (Miq.) Miq. ex Havil. is a famous traditional Chinese medicine used in managing hyperpyrexia, epilepsy, preeclampsia, and hypertension in China. Hypothesis/purpose: To discover the potential natural human carboxylesterase 2 (hCE 2) inhibitors from U. rhynchophylla. Methods: Compounds were obtained from the hooks of U. rhynchophylla by silica gel and preparative HPLC. Their structures were elucidated by using HRESIMS, 1D and 2D NMR spectra. Their inhibitory activeties and inhibition kinetics against hCE 2 were assayed by the fluorescent probe, and potential mechanisms were also investigated by molecular docking. Results: Twenty-three compounds, including a new phenolic acid uncariarhyine A (1), eight known triterpenoids (2-9), and ten known aromatic derivatives (10, 13-16, and 19-23), were isolated from U. rhynchophylla. Compounds 1-5, 7, 9, and 15 showed significant inhibitory activities against hCE 2 with IC50 values from 4.01 +/- 0.61 mu M to 18.60 +/- 0.21 mu M, and their inhibition kinetic analysis results revealed that compounds 1, 5, 9, and 15 were non-competitive; compounds 3 and 4 were mixed-type, and compounds 2 and 7 were uncompetitive. Molecular docking studies indicated inhibition mechanisms of compounds 1-5, 7, 9, and 15 against hCE 2. Conclusion: Our present findings highlight potential natural hCE 2 inhibitors from U. rhynchophylla.

Tissue adhesives play an important role in clinical applications and may aid in the treatment of cartilage defects for improved cartilage integration. However, many fail to satisfy the demand for adequate adhesive strength on wet tissue surfaces and to facilitate sufficient cell migration and extracellular matrix (ECM) deposition at the defect site. Utilizing poly(2-alkyl-2-oxazoline) (POx)-based polymers equipped with mussel-inspired adhesion moieties and combining them with the natural wound sealant fibrinogen, we fabricated an adhesive biosynthethic hydrogel with tunable mechanical properties and improved bonding strength. Degradation of the hydrogels could be adjusted by the ratio of amide to ester linkages of the catecholic functional group at the POx side chain. In an in vitro disc/ring model for lateral cartilage integration, a benefit in long-term integration was observed with enhanced degradation of the adhesive without the expense of bonding strength. Incorporation of degradable ester linkages in the polymer facilitated cell invasion and strong deposition of cartilaginous ECM at the defect site. Overall, the results suggest that the presented injectable adhesive hydrogel, due to its easy tunability, holds great potential for cartilage defect treatment and other medical applications. It is textbook knowledge that carboxamides benefit from increased stabilisation of the electrophilic carbonyl carbon when compared to other carbonyl and carboxyl derivatives. This results in a considerably reduced reactivity towards nucleophiles. Accordingly, a perception has been developed of amides as significantly less useful functional handles than their ester and acid chloride counterparts. However, a significant body of research on the selective activation of amides to achieve powerful transformations under mild conditions has emerged over the past decades. This review article aims at placing electrophilic amide activation in both a historical context and in that of natural product synthesis, highlighting the synthetic applications and the potential of this approach.
