Antibiotics in soil environments are a growing concern. Identifying transformation products is key to elucidating degradation pathways and mechanisms of antibiotics and other organic micropollutants. The primary challenge of transformation product identification is the interference of matrices. In this study, stable-isotope assisted nontarget screening was used to identify biodegradation products of florfenicol in soil. A total of 74 candidates were prioritized from thousands of mass features observed by a tiered peak filtering approach. Moreover, with the support of in silico prediction tools, the structures of 12 transformation products were elucidated, and 9 of them were reported for the first time. A biodegradation map of florfenicol consisting of amide hydrolysis, dechlorination, dehydration, defluorination, and sulfone reduction was established based on these identified products. A total of 8 products were also found in 6 field soil samples with manure application. Because of the structural similarity to florfenicol, some transformation products might still keep antimicrobial activity toward a variety of bacterial species. The strategies demonstrated in this study provide a basis for efficient identification of transformation products of other organic micropollutants in a variety of environmental matrices. The results also shed light on the degradation mechanisms, risk assessments, and regulations of these compounds.

The fatty acid amide hydrolase (FAAH) gene was involved in the modulation of reward and addiction pathophysiology of illicit drugs abuse, and its polymorphisms might be associated with risk of methamphetamine (METH) dependence. This study aimed to investigate the FAAH mRNA levels in peripheral blood mononuclear cells and plasma protein levels and to analyze the 385C/A polymorphism (rs324420) between METH-dependent patients and controls. The levels of FAAH mRNA in METH dependence were significantly lower than in controls (P < 0.001), however, its plasma protein underwent a significant similar to 2-fold increase (P < 0.001). The A allele of the 385C/A polymorphism significantly increased the METH dependence risk (P < 0.001, odds ratio [OR] = 1.646, 95% confidence interval [CI] = 1.332-2.034). The carried A genotypes (AA, AC, and AA/AC) of 385C/A polymorphism also increased METH-dependence risks under a different genetic model (AA vs. CC: P = 0.017, OR = 2.454, 95%CI = 1.171-2.143; AC vs. CC: P < 0.001, OR = 1.818, 95%CI = 1.404-2.353; AC/AA vs. CC: P < 0.001, OR = 1.858, 95%CI = 1.444-2.319). The similar results were obtained after adjusting for age and sex. Unfortunately, we failed to find that any genotype of 385C/A polymorphism affected the mRNA or plasma protein levels in controls, respectively (P > 0.05). These data indicate that the FAAH may play an important role in the pathophysiological process of METH dependence, and the 385C/A polymorphism may be associated with METH dependence susceptibility in a Chinese Han population.

Here, we used selective pharmacological compounds during operant training to determine the impact of fatty acid amide hydrolase (FAAH) inhibition to increase anandamide (and other n-acylethanolamines) or monoacylglycerol lipase (MAGL) inhibition to increase 2-AG levels on the formation of habitual behaviors in mice using a food-reinforced contingency degradation procedure. We found, contrary to our hypothesis, that inhibition of FAAH and of MAGL disrupted the formation of habits. Next, AM251 was administered during training to verify that impaired habit formation could be assessed using contingency degradation. AM251-exposed mice responded at lower rates during training and at higher rates in the test. To understand the inconsistency with published data, we performed a proof-of-principle dose-response experiment to compare AM251 in our vehicle-solution to the published vehicle-suspension on response rates. We found consistent reductions in response rate with increasing doses of AM251 in solution and an inconsistent dose-response relationship with AM251 in suspension. Together, our data suggest that further characterization of the role of CB1R signaling in the formation of habitual responding is warranted and that augmenting endocannabinoids may have clinical utility for prophylactically preventing aberrant habit formation such as that hypothesized to occur in substance use disorders.

Mechanistic studies, based on theoretical calculations, indicate that the CAAC ligand plays an important role in promoting polarity reversal of hydride of HBpin; it serves as an H-shuttle to facilitate deoxygenative hydroboration. The preparation of several commercially available pharmaceuticals by means of this strategy highlights its potential application in medicinal chemistry.

The low resilience and poor elongation characteristics of thermoplastic polyurethane (TPU) foams make it unsuitable for applications such as high-end sports shoes. In this work the incorporation of thermoplastic amide elastomer (TPAE) into TPU foam has been investigated to meet address this shortcoming. Five TPU/TPAE composite pellets were first prepared using varying combinations of TPU, compatibilizer additive, antioxidant, processing aid, and UV stabilizer. Out of these studies, the pellet with the most hardness was foamed with supercritical CO2, to obtain the TPU/TPAE composite foams. The TPU/TPAE composite foams consisting of aminized polyether polyol, nylon 6 (as the hard segment), and Jeffamine D400 (as the soft segment) exhibit excellent resilience, high elongation, good hardness, high tensile strength, and smaller cell size. Experimental results demonstrated that introduction of TPAE into the TPU composite foam efficiently improves their mechanical performance.

In this work, poly(methacrylated vanillin) (PMV) was investigated for its reactivity in multicomponent reactions as a reactive polymer that can be sourced from lignin-based components. Solid-phase extraction with an Ultra Performance Liquid Chromatograph-Mass Spectrometer in electrospray ionization mixed mode (5-fluorouracil and bicalutamide in negative mode and the others in positive one) was developed to determine seven cytostatics in wastewater and surface water. Among eight tested cartridges with different sorbents and conditions, the best extraction performance was attained with Oasis WAX at pH 10, with recoveries ranging from 31 +/- 4 (5FU) and 103 +/- 17% (MPA). Regarding the chromatographic analysis, the best results were achieved with an XBridge amide column.
