beta-N-Acetylhexosaminidases are widely distributed exoglycosidases and have attracted significant attention due to their important roles in the field of pesticide and drug discovery. Remarkably, human O-GlcNAcase (hOGA) and human beta-N-acetylhexosaminidase (HsHex) possess the same catalytic mechanism but play different physiological actions in vivo. In this Letter, we aim to improve the inhibitory potency and selectivity of previously reported thioglycosyl-naphthalimides against hOGA. The rational compound design led to the synthesis of 13r bearing a 4-piperidylnaphthalimide moiety as a highly potent hOGA inhibitor (K-i = 0.6 mu M against hOGA) with good selectivity (K-i > 100 mu M against HsHexB). Furthermore, to investigate the basis for the potency and selectivity of 13r against hOGA, the possible inhibitory mechanisms of selected inhibitors (15b, 13b, and 13r) against hOGA and HsHexB were studied using molecular docking and MD simulations. These 4-substituted naphthalimide thioglycosides may potentially serve as useful tools for the further study of the function of hOGA.

Rapid and sensitive detection technology is the key to preventing food-borne disease outbreaks. In this study, a low-field nuclear magnetic resonance (NMR) biosensor based on polyamidoamine dendrimers was prepared for the rapid detection of Salmonella in milk. The polyamidoamine dendrimer was biotinylated by amide reaction and chelated to diethylene triamine pentacetate acid and gadolinium to form magnetic complexes. The antibody and magnetic complexes were combined through a streptavidin-biotin system using streptavidin as an intermediate bridge to obtain the immunoprobe. Salmonella was captured by the irnmunoprobe via antigen-antibody interaction and then separated from the mixture by membrane filtration. Finally, the longitudinal relaxation signal of the filtrate was obtained by NMR. The biosensor had excellent anti-interference capability and could detect Salmonella within 1.5 h at a sensitivity of 10(3) cfu mL(-1). This method based on NMR can realize detection in complex samples and has the potential to be a quick and nondestructive method for detecting target bacteria.

Degradable polymer prodrugs based on gemcitabine (Gem) as an anticancer drug were synthesized by drug-initiated’ nitroxide-mediated radical ring-opening copolymerization (NMrROP) of methacrylic esters and 2-methylene-4-phenyl-1,3-dioxolane (MPDL). Different structural parameters were varied to determine the best biological performances: the nature of the monomer [i.e., oligo(ethylene glycol) methacrylate (OEGMA) or methyl methacrylate (MMA)], the nature of the Gem-polymer linker (i.e., amide or amide and diglycolate) and the MPDL content in the copolymer. Depending on the nature of the methacrylate monomer, two small libraries of water-soluble copolymer prodrugs and nanoparticles were obtained (M-n approximate to 10000 g mol(-1), D = 1.1-1.5), which exhibited tunable hydrolytic degradation under accelerated conditions governed by the MPDL content. Drug-release profiles in human serum and in vitro anticancer activity on different cell lines enabled preliminary structure-activity relationships to be established. The cytotoxicity was independently governed by: (i) the MPDL content – the lower the MPDL content, the greater the cytotoxicity; (ii) the nature of the linker – the presence of a labile diglycolate linker enabled a greater Gem release compared to a simple amide bond and (iii) the hydrophilicity of the methacrylate monomer-OEGMA enabled a greater anticancer activity to be obtained compared to MMA-based polymer prodrugs. Remarkably, the optimal structural parameters enabled reaching the cytotoxic activity of the parent (free) drug.

This research offers a theoretical foundation for the design and development of novel composite food gels with various functional properties.

N-[difluoro(methyl)silyl]carboxamides: Synthesis, structural features and theoretical estimating of Si <- O dative bond energy The new original method of the synthesis of N-[difluoro(methyl)silyl]methylcarboxamides and related compounds based on the reaction of alkylation of N-trimethylsilyl derivatives by bifunctional silane ClCH2SiF2Me was use to prepare of N-{[difluoro(methyl)silyl]methyl}-N-methylacetamide, N-{[difluoro-(methyl)silyl]methyl}benzanilide and phenyl-N-phenyl-N-{[difluoro(methyl)silyl]methyl}carbamate. The structures of these compounds confirmed by NMR, FTIR and X-ray analysis. According to X-ray analysis data the lengths of coordination bonds C=O -> Si in studied compounds are 1.969, 2.000 and 2.176 angstrom respectively. The lengths of Si-F-ax. comprise 1.686, 1.661 and 1.632 angstrom respectively. QTAIM analysis was used to estimate the energy of co-ordination bond C=O -> Si in these amides.
