Two peptide nucleic acid (PNA) oligomers containing two different organometallic moieties, namely derivatives of azidoferrocene (Fc-N3) and ferrocene carboxylic acid (Fc-COOH) for PNA5 and derivatives of β-cymantrenoylpropionic acid [Cym-CO(CH2)2COOH] and the rhenium bisquinoline tricarbonyl complex [Re(BQ-N3)(CO)3]Br for PNA6, have been prepared on a solid support by using two different synthetic methods: Peptide coupling and click chem. PNA5 and PNA6 were unambiguously characterized by ESI-MS or MALDI-TOF MS and their purity checked by LC-MS. As expected, the bioconjugate PNA6 presents two broad and strong absorption bands at 1933 and 2032 cm-1 in its IR spectrum due to the presence of six metal-carbonyl bonds. It has also been demonstrated that the presence of the Mn complex in PNA6 did not significantly alter the fluorescence properties of the Re complex in aqueous solution compared with a previously reported Re-PNA bioconjugate (PNA7). Photoexcitation of PNA6 at 350 nm reveals two distinct emission bands at about 434 and 595 nm in aqueous solutions PNA6 was successfully electroporated into HeLa cells, as shown by high-resolution continuum-source at. absorption spectroscopy, which was used to measure the concentrations of both Re and Mn. However, no fluorescence of the Re complex in PNA6 was observed in living cells, even at a concentration 20 times higher than that previously reported for PNA7.

Preliminary microrheol. experiments were performed on both systems to evaluate their viscoelastic properties which resulted in agreement with the formation of soluble hyperbranched polymers that could be useful for drug/gene delivery, as well as for encapsulating organic pollutants of different shapes and sizes in environmental applications. Double-helical RNA has become an attractive target for mol. recognition because many noncoding RNAs play important roles in the control of gene expression. Recently, we discovered that short peptide nucleic acids (PNA) bind strongly and sequence selectively to a homopurine tract of double-helical RNA via formation of a triple helix. Herein, we tested if the mol. recognition of RNA could be enhanced by α-guanidine modification of PNA. Our study was motivated by the discovery of Ly and co-workers that the guanidine modification greatly enhances the cellular delivery of PNA. Isothermal titration calorimetry showed that the guanidine-modified PNA (GPNA) had reduced affinity and sequence selectivity for triple-helical recognition of RNA. The data suggested that in contrast to unmodified PNA, which formed a 1:1 PNA-RNA triple helix, GPNA preferred a 2:1 GPNA-RNA triplex invasion complex. Nevertheless, promising results were obtained for recognition of biol. relevant double-helical RNA. Consistent with enhanced strand invasion ability, GPNA derived from D-arginine recognized the transactivation response element of HIV-1 with high affinity and sequence selectivity, presumably via Watson-Crick duplex formation. On the other hand, strong and sequence selective triple helixes were formed by unmodified and nucleobase-modified PNA and the purine-rich strand of the bacterial A-site. These results suggest that appropriate chem. modifications of PNA may enhance mol. recognition of complex noncoding RNAs.

Although peptide nucleic acids (PNAs) have improved nuclease resistance compared with DNA or RNA, it is difficult to synthesize long PNAs because of poor elongation yield. Herein we synthesized 20-mer PNAs (PNA20), targeting Nnmt mRNA, as well as its conjugate with testosterone 17β-carboxylic acid, in high purity and yield. This synthesis was conducted using Oxyma as a condensation agent and NMP as a solvent for Fmoc-PNA-C(Bhoc)-OH. The resistance of PNA20 to exonuclease was higher than that of RNA. Furthermore, the abilities of PNA20 and its conjugate to bind to complementary DNA were stronger than that of DNA or RNA. These findings lay the basis for the synthesis of long PNA derivatives toward oligonucleotide therapeutics.

A novel, cationic cysteine-rich CPP, CyLoP-1, has been developed exhibiting distinguished diffused cytosolic distribution along with endosomal uptake at low micromolar concentrations Comparative uptake anal. with known CPPs showed CyLoP-1 as a promising delivery vector to access the cytosol in a variety of cell types. In addition to the pos. charged residues, the presence of cysteines and tryptophans proved to be essential to maintain its functionality. Also, the oxidation status of the cysteines played an important role for the uptake efficiency of CyLoP-1, with the disulfide-containing form being more effective. The distinct feature of CyLoP-1 to enter the cytosol was further explored by the covalent attachment of cargoes of different nature and sizes. In particular, induction of caspase-3 activity (indicating apoptosis) by a CyLoP-1-SmacN7 conjugate proved successful delivery of the pro-apoptotic cargo to its site of action in the cytosol. Efficient intracellular delivery into the entire cytosol already at low micromolar concentrations makes CyLoP-1 a promising candidate for cytosolic delivery of cargoes of small sizes. Thus, this peptide might prove to be useful for efficient transmembrane delivery of agents directed to cytosolic targets.

A novel simplified model was introduced for membrane fusion mediated by SNARE proteins.
