The brain’s endocannabinoid system plays a crucial role in reward processes by mediating appetitive learning and encoding the reinforcing properties of substances. Evidence also suggests that endocannabinoids are an important constituent of neuronal substrates involved in emotional responses to stress. We found that proxies for endocannabinoid system signaling (i.e., endocannabinoid genes and chronic exposure to cannabis) and stress have differential effects on neural response to cannabis cues. Specifically, a single nucleotide polymorphism (SNP) variant in the cannabinoid receptor 1 (CNR1) gene, early life stress, and current perceived stress modulated reward responsivity in long-term, heavy cannabis users, while a variant in the fatty acid amide hydrolase (FAAH) gene and current perceived stress modulated cue-elicited response in non-using controls. These associations were related to distinct neural responses to cannabis-related cues compared to natural reward cues. Understanding the contributions of endocannabinoid system factors and stress that lead to downstream effects on neural mechanisms underlying sensitivity to rewards, such as cannabis, will contribute towards a better understanding of endocannabinoid-targeted therapies as well as individual risks for cannabis use disorder.

In this study, we examine the influence of three aqueous metal iodide salt solutions (LiI, NaL and CsI) on the phase transition temperature of poly(N-isopropylacrylamide) (PNIPAM) by measuring the turbidity change of the solutions. Weakly hydrated anions, such as iodide, are known to interact with the polymer and thereby lead to salting-in behavior at low salt concentration followed by salting-out behavior at higher salt concentration. When varying the cation type, an unexpected salting-out trend is observed at higher salt concentrations, Cs+ > Na+ > Li+. Using molecular dynamics simulations, it is demonstrated that this originates from contact ion pair formation in the bulk solution, which introduces a competition for iodide ions between the polymer and cations. The weakly hydrated cation, Cs+, forms contact ion pairs with I- in the bulk solution, leading to depletion of CsI from the polymer-water interface. Microscopically, this is correlated with the repulsion of iodide ions from the amide moiety.

The present work describes a highly efficient synthetic strategy for amides via oxidative coupling of benzaldehydes or benzylamines with N-substituted formamides using a heterogeneous Co/Al hydrotalcite-derived catalyst in the presence of TBHP. Preparation and characterization of pickering emulsion stabilized by hordein-chitosan complex particles

In this study, hordein-chitosan complex particles (HCPs) were prepared using anti-solvent precipitation method. The addition of chitosan led to the increase in the particle diameter and zeta-potential as well as the gradual decrease in fluorescence intensity values of HCPs, indicating chitosan absorbed onto the surface of hordein particles. Fourier transform infrared spectra of HCPs showed the significant shifts of peak position for O-H stretching vibration, amide I and II bands after addition of chitosan, suggesting the formation of hydrogen bonds and hydrophobic interactions between hordein and chitosan. Three-phase contact angle (theta(o/w)) of HCPs revealed the excellent wettability (theta(o/w) = 89.3 +/- 1 degrees) of particles when the mass ratio of hordein to chitosan was 2:1 (HCPs(2:1)). Oil-in-water Pickering emulsions (Caprylic/Capric Triacylglyceride as oil phase) stabilized by HCPs(2:1) were further prepared. Confocal laser scanning microscope images indicated that HCPs absorbed at the oil/water interface and formed a dense steric barrier on the surface of spherical oil droplets. Higher oil fractions (50% and 60%) led to the formation of stable Pickering emulsion gels, which showed long-term storage stability. This study provides a new solution to accelerate the application of hordein-based Pickering emulsions in food industry.

Synthesis and characterization of thermo-sensitive block copolymer of N-isopropyl acryl amide-b-Alanine [(NIPAM)-b-(Alanine)] thin film and its doping with (AuNPs)-(SiO2NPs), (gold and silica nanoparticles) were reported. Further, composite effect on K562 (leukemia) cells was examined based on in vitro cell-based studies. The synthesis of SiO2NPs was followed through facile Stober’s sol-gel synthesis methods. The individual morphology of [(NIPAM)-b( Alanine)] thin film, AuNPs and SiO2NPs including [(NIPAM)-b-(Alanine)]@(Au)-(SiO2NPs) composite was confirmed by using TEM instrumentation. [(NIPAM)-b-(Alanine)] thin film was embedded with gold and silica nanoparticles followed by the sonication. The average size of AuNPs is 16 nm and for SiO2NPs, it is 368 nm (in diameter). Synthesized composite [(NIPAM)-b-(Alanine)]@(Au)@(SiO2NPs) is biocompatible for mankind use. However, composite used to examine the inhibitory activity on K562 cells and it shows 78% inhibition, which is significant value for 24 h treatment under humidified atmospheric conditions.

We report the diastereoselective synthesis of novel spiropiperidine templates for use in SAR studies of beta-secretase (BACE) inhibitors and also as versatile ligands for other receptor types. The overall synthetic approach stems from chiral starting material benzyl (S)-2-methyl-4-oxopiperidine-1-carboxylate and employs an Overman rearrangement to control the stereochemistry at the quaternary center. This process is followed by a Grubbs metathesis to close a five-membered top ring to form an alpha,beta-unsaturated lactam or an alpha,beta-unsaturated sultam. We also demonstrate that this chemistry can accommodate additional substituents on the lactam/sultam ring and allows late stage sequential functionalization of the amine and amide nitrogens to rapidly produce diverse analogues.

To adapt to the environment, it is important for insects to learn and evaluate food quality to avoid toxins in food. Currently, attractants are widely used for pest control, as they are environmentally friendly pesticides. Learning to associate an attractive odor with a toxin in food may influence insect behaviour in response to the attractant, thus affecting its application. In this study, we found that Bactrocera dorsalis, a serious pest of fruits and vegetables, can learn to associate a male lure compound, methyl eugenol (ME), with a sucrose solution contaminated with N,N-diethyl-3-methyl benzoyl amide (DEET), a common insect repellent and decrease its proboscis extension response (PER) elicited by ME. Learning with DEET depends on its concentration, and flies exhibited significant aversive memory formation when high concentrations were used. Aversive-DEET memory formed with 0.6% DEET persisted for at least 24hr. We also found that aversive memory formed during training could be transferred to a behavioural response in an operant context.
