The ability of the 2-substituted aniline motif to serve as a scaffold for designing potential LuxR-regulated quorum sensing (QS) modulators has been investigated, using docking experiments and biological evaluation of a series of 15 specially synthesized compounds. Aniline, 2-acetyl-aniline and 2-nitroaniline were considered, as well as their N-acylated derivatives. Docking experiments showed that the 2-substituted aniline motif fits within the LuxR binding site at the place of the lactone moiety of AHL, and the biological evaluation revealed QS antagonisitic activity for several compounds, validating the hypothesis that this scaffold acts on QS. Structure activity relationships are discussed regarding interactions with the key residues of the LuxR binding site, showing significant variations in the H-bonding pattern.

Opioid use for chronic pain is limited by severe central adverse effects. We examined whether activating mu-opioid receptors (MORs) in the peripheral nervous system attenuates spinal cord injury (SCI) pain-like behavior in mice. We produced a contusive SCI at the T10 vertebral level and examined motor and sensory dysfunction for 6 weeks. At 6 weeks, we tested the effect of subcutaneous (s.c.) injection of dermorphin [D-Arg2, Lys4] (1 -4) amide (DALDA), a peripherally acting MOR-preferring agonist, on mechanical and heat hypersensitivity. Basso mouse scale score was significantly decreased after SCI, and mice showed hypersensitivity to mechanical and heat stimulation at the hind paw beginning at 2 weeks, as indicated by increased paw withdrawal frequency to mechanical stimulation and decreased paw withdrawal latency to heat stimulation. In wild-type SCI mice, DALDA (1 mg/kg, s.c.) attenuated heat but not mechanical hypersensitivity. The effect was blocked by pretreatment with an intraperitoneal injection of methylnaltrexone (5 mg/kg), a peripherally restricted opioid receptor antagonist, and was also diminished in Pirt-MOR conditional knockout mice. DALDA did not adversely affect exploratory activity or induced preference to drug treatment in SCI mice. In vivo calcium imaging showed that DALDA (1, 10 mg/kg, s.c.) inhibited responses of small dorsal root ganglion neurons to noxious heat stimulation in Pirt-GCaMP6s mice after SCI. Western blot analysis showed upregulation of MOR in the lumbar spinal cord and sciatic nerves at 6 weeks after SCI. Our findings suggest that peripherally acting MOR agonist may inhibit heat hypersensitivity below the injury level with minimal adverse effects.

This study demonstrated an efficient activation of Oxone by ultraviolet light A-irradiated Fe-III-nitrilotriacetate complex to induce the generation of sulfate and hydroxyl radicals (i.e., SO4 center dot- and HO center dot) under initial neutral pH. The important parameters such as the solution pH, the molar ratio of nitrilotriacetate:Fe-III, the dosages of Oxone and Fe-III-nitrilotriacetate complex were evaluated in terms of the degradation kinetics of an emerging contaminant Crotamiton. The results indicated that fast degradation rates of crotamiton were achieved under initial circumneutral conditions (e.g., pH 5.0-7.0), with apparent rate constants at 0.0936-0.1287 min(-1) (the ultraviolet light fluence-based rate constants at 0.48-0.66 J(-1) cm(2)). In addition, the optimal molar ratio of nitrilotriacetate:Fe-III was determined as 1:1, larger ratios decreased the degradation rate of crotamiton due to the competition effect of nitrilotriacetate on SO4 center dot-. The suitable dosages of Oxone and Fe-III-nitrilotriacetate complex were determined as 0.5 mM and 0.1 mM, respectively. Under the given optimal conditions, more than 99% degradation efficiency of crotamiton was achieved at an ultraviolet light fluence of 3.90 J cm(-2), better than those results obtained by the activation of S2O82- and H2O2. The results of quenching tests (tert-butyl alcohol and 2-propanol as scavengers) suggested that SO4 center dot- and HO center dot contributed similar to 65% and 35% to the degradation of crotamiton, respectively. Furthermore, the identified intermediates includes hydroxy-crotamiton, dihydroxy-crotamiton, aldehyde-crotamiton, aldehyde-dihydroxy-crotamiton, N-ethyl-2,3-dihydroxy-N-(o-tolyl)butanamide, (E)-N-ethyl-N-phenylbut-2-enamide, 2-(ethylamino)benzaldehyde and/or 2-(o-tolylamino)acetaldehyde, and (E)-N-ethylbut-2-enamide. The results indicated that the SO4 center dot- preferentially attacked on the amide and methyl groups of crotamiton. This work provided insight into the efficacy, radicals formation and mechanism on the activation of Oxone by ultraviolet light A-irradiated FeIII-nitrilotriacetate complex, offering an alternative approach for advanced water treatment.

Mass-selective two-color resonant two-photon ionization (2C-R2PI), UV/UV hole-burning, and infrared (IR) depletion spectra of supersonic jet-cooled 2-pyridone center dot(H2O)(n) clusters with n = 1-4 have been measured to investigate the local hydration patterns around 2-pyridone (2PY) as a function of cluster size. As shown by others, the IR frequencies of the OH and NH stretches of the n = 1, 2 clusters are characteristic of water wires stretching from the NH to the C=O group of 2PY. We identify two isomers (3A and 3B) of the n = 3 cluster in the 2C-R2PI spectrum and separate them by IR/UV and UV/UV hole-burning techniques. Isomer 3A exhibits a three-membered water wire, extending the n = 1, 2 structural motif. Isomer 3B exhibits bifurcated water wires with the first H2O donating to two waters that form H-bonds to the C=O group. This increases the H-bond strength between the NH group of 2PY and the proximal H2O molecule, lowering the NH stretch to similar to 2800 cm(-1) The n = 4 cluster is also bifurcated with two water wires between the bifurcating H2O and the C=O group. The cluster-selective IR spectra are complemented with density-functional calculations using the PW91, B3LYP, B97-D, and M06-2X functionals, where the latter two include long-range dispersive interactions, and with the ab initio correlated SCS-CC2 method. The calculated IR spectra provide firm assignments of the structures of the n = 1-4 cluster structures and allow us to understand the evolution of individual H-bond strengths with increasing cluster size.

In the present research article, a hyperbranched alkyd resin was synthesized by using bio-based material (castor oil based fatty amide) and pyromellitic dianhydride. The synthesized hyperbranched alkyd resin was characterized by nuclear magnetic resonance (NMR) and Fourier transform infrared (FTIR) spectroscopy. The polyurethanes (PMFAI and PMFAM) were prepared from hyperbranched alkyd resin and diisocyanates [isophorone diisocyanates (IPDI) and methylene diphenyl diisocyanate (MDI)] maintaining the OH:NCO ratio of 1:1.2. These polyurethanes were used in coating applications on mild steel and wood panels. The performance of cured polyurethane coatings was studied in terms of crosscut adhesion, impact resistance, gloss, scratch hardness, and chemical resistance. The chemical resistance test was performed by immersion method in 2 N HCl, 2 N NaOH, 3.5% NaCl solution, xylene, and water. The corrosion rate and anticorrosion efficiency of the coating panels were determined by a potentiostat. From a study of coating properties, it was found that the PMFAI and PMFAM exhibited better coating properties as compared to reported linear alkyd polyurethanes. The stain test of coatings was performed on wood panels. The thermal behavior of the coatings was determined by thermogravimetric analyzer. The surface morphology of coating films was examined by scanning electron microscopy and atomic force microscopy. It was found that the coating properties of hyperbranched alkyd resin were excellent as compared to linear alkyd resin polyurethanes.

Subsequently amide groups present in the PAMAM have been reduced using lithium aluminum hydride to yield polyamine dendrimer namely poly (N-propylethane-1,2-diamine) G(3). Further, peripheral amine groups were grafted with succinic anhydride to achieve carboxylic acid terminated dendrimer. The resulting nanomaterial was confirmed by FTIR, XRD, VSM, TGA, HRTEM and conductometry methods. In order to emphasize the structural efficacy of polyamine dendrimer, the metal ion uptake behavior has been discussed for the representative cations such as Pb2+, Cu2+, Cr6+, Ca2+ and Ni2+ under the environmental concern.
