Structural variations of the benzylphenoxyacetamide (BPA) molecular skeleton were explored as a viable starting point for designing new anti-glioblastoma drug candidates. Hand-to-hand computational evaluation, chemical modifications, and cell viability testing were performed to explore the importance of some of the structural properties in order to generate, retain, and improve desired anti-glioblastoma characteristics. It was demonstrated that several structural features are required to retain the anti-glioblastoma activity, including a carbonyl group of the benzophenone moiety, as well as 4′-chloro and 2,2-dimethy substituents. In addition, the structure of the amide moiety can be modified in such a way that desirable anti-glioblastoma and physical properties can be improved. Via these structural modifications, more than 50 compounds were prepared and tested for anti-glioblastoma activity. Four compounds were identified (HR28, HR32, HR37, and HR46) that in addition to HR40 (PP1) from our previous study, have been determined to have desirable physical and biological properties. These include high glioblastoma cytotoxicity at low mu M concentrations, improved water solubility, and the ability to penetrate the blood brain barrier (BBB), which indicate a potential for becoming a new class of anti-glioblastoma drugs.

Insect-microbial symbioses have vast biochemical diversity, which is beneficial to produce bioactive secondary metabolites. In this study, chemical examination of a Streptomyces sp. associated with a mud dauber wasp led to the isolation of fourteen compounds. Their structures were determined by spectroscopic methods and comparison with literature data. Among the isolates, compounds 1,2,3-benzotriazin-4(1H)-one and 4-(2-aminoethyl)phenyl acetate were first reported from this species. Bioactivities of the isolated compounds were assayed for the first time against hexokinase II. 4-(2-Aminoethyl)phenyl acetate, germicidin B, phenylacetic acid, isogermicidin A and germicidin C displayed significant inhibitory activity against hexokinase II, with the IC50 values of 5.11, 7.11, 7.15, 8.45 and 8.78 mu M, respectively.

A novel Pd/Cu-cocatalyzed carbonylative cyclization by C-H activation and N-dealkylative C-N bond activation has been developed for the chemoselective construction of synthetically useful heterocycles. The N,N-dimethylamine group on o-indolyl-N,N-dimethylarylamines was found to act as both the directing group and reactive component in this C-H carbonylative cyclization reaction. Furthermore, a unique C-H oxidation/carbonylative lactonization of diarylmethylamines is firstly demonstrated under modified reaction conditions, which could be easily applicable to the one-step synthesis of multi-substituted phthalides bearing an N,O-ketal skeleton that is difficult to access by previously reported methods. Mechanistic studies implicate that Pd/Cu-cocatalyzed C-H oxidation/carbonylative lactonization is a sequential reaction system via Cu-catalyzed C(sp(3))-H oxidation and Pd-catalyzed oxidative carbonylation of the C(sp(2))-H bond. It was found that trace amounts of water are essential to promote the Cu-catalyzed C(sp(3))-H oxidation of diarylmethylamine for the formation of the hydroxyl group, which could act as an in situ-formed directing group in the intramolecular carbonylative lactonization step.

Metal-free, visible-light-induced denitrogenative phosphorylation of 1,2,3-benzotriazinones was achieved. With the use of eosin Y as a photoredox catalyst, N,N-diisopropylethylamine as a base, CH3CN-H2O as a solvent and sunlight or a blue LED as a light source, a variety of aryl-phosphonates, aryl-phosphinates, and aryl-phosphine oxides were efficiently prepared. In addition, B(2)pin(2) instead of P-nucleophiles as a radical acceptor was also demonstrated. The key advantages of this newly developed method are the clean reaction profile, use of a low-cost organic-dye catalyst, energy efficiency, broad substrate scope, good to excellent yields and large-scale synthetic applicability. The gram-scale synthesised compounds could be isolated in pure form just upon extraction, followed by re-crystallisation; no tedious chromatographic purification was required.

A visible light-induced and exogenous photo-sensitizer-free, copper-catalyzed radical aminocarbonylation of cycloketone oxime esters with CO gas and amines is developed. The mild catalytic system involving CuCl and N,N,N-tridentate ligand shows good reactivity and chemo-selectivity, and tolerates a wide range of cycloketone oxime esters and alkyl/aryl amines, giving the corresponding cyanoalkylated amides in moderate to good yields. Preliminary mechanistic studies suggest that the reaction involves a Cu-I/(CuCuIII)-Cu-II/-based catalytic cycle and radical intermediate.

A simple and efficient protocol for the formylation of amines with formic acid, catalyzed by a polyoxometalate-based chromium catalyst, is described. Notably, this method shows excellent activity and chemo-selectivity for the formylation of primary amines; diamines have also been successfully employed. Importantly, the chromium catalyst is potentially non-toxic, environmentally benign and safer than the widely used high valence chromium catalysts such as CrO3 and K2Cr2O7. The catalyst can be recycled several times with a negligible impact on activity. Finally, a plausible mechanism is provided based on the observation of intermediate and control experiments.

A new metal-free and modular approach for the synthesis of various functionalized dihydroquinazolinones has been developed from isatoic anhydride, amines, 4-chloro-N-hydroxybenzimidoylchloride to yield up to 71%. The reaction has been screened in various bases, solvents at different temperatures. The substrate scope of the reaction has been studied with various amines and the possible reaction mechanism for this reaction has also been proposed.

In this paper, we report on a new dielectric, a C-60-containing block polyimide (PI-b-C-60). This was realized by introducing C-60 as pendent groups into a polymer backbone. When this dielectric was used in a non-contact mode triboelectric nanogenerator (TENG), it achieved high output power and reliable operation. Compared with perfluoroalkoxy alkane film-based TENGs, the TENG based on PI-b-C-60 generated 4.3 times higher output power and a superior charge density of over 300 mu C m(-2) with a 3 times slower charge decay rate. These results are most likely due to the excellent charge-retention characteristics induced by the most negative electrostatic potential of C-60 within the backbone, and these characteristics were confirmed by surface potential measurements. Furthermore, in the course of our work, two non-contact mode applications, a keyless electronic door lock system and a speed sensor with a tone wheel for a car, were developed. Without an ion injection process being needed, very sensitive and reliable operations of the speed sensor were successfully demonstrated, even under very humid conditions (similar to 99% RH).
